In November 2009 I had my first PCR test done with a result of 0.175 then in april 0.150 and now in August 2010 it is 0.125.  not sure what this means.  I had the test done at LabCorp and these are the results my doctor told me he got.  He wasn't much help explaining.  I was diaganosed in May 2009 and FISH was down from 200 to 2 by November after 400 mg of Gleevec started in May.  That was when we switched to PCR test.  Doctor is very happy with results but I am wondering since it doesn't look like any log reduction since pcr started last november.  doc has increase dosage, after I asked about log reduction, to 800 mg on day and 400 mg next day for the gleevec.

should I have expected 0.175 then 0.0175 the 0,00175 as a three log reduction.  not sure what scale only that LabCorp is doing the testing.

The FISH shows a good response, which is important information.  The PCRs are flat, assuming LabCorp has not changed reporting methods (I say this because some labs are switching to International Scale, and LabCorp had not swicthed last I knew).  Assuming the PCRs reflect a "plateau" in your response to drug therapy, which it seems to, then you might want to talk with your Onc about either increasing Gleevec dosage or switching to another drug.  It is a very good idea to keep pushing the leukemia levels downward.  If a plateau occurs at or below a 3 log reduction, then that is probably just fine.  But you are probably at a roughly 2 log reduction since you are close to Complete Cytogenetic Response (zero FISH will be CCR).

How can FISH be greater than 100?

Diagnosed 11 May 2011 (100% FiSH, 155% PCR)

with b2a2 BCR-ABL fusion transcript coding for the 210kDa BCR-ABL protein

Sprycel: 20 mg per day - taken at lights out with Quercetin and/or Magnesium Taurate

6-8 grams Curcumin C3 complex.

2015 PCR: < 0.01% (M.D. Anderson scale)

2016 PCR: < 0.01% (M.D. Anderson scale)

March        2017 PCR:     0.01% (M.D. Anderson scale)

June          2017 PCR:     "undetected"

September 2017 PCR:     "undetected"

I believe they count 200 cells and see how many are leukemic.

None of the numbers used by anyone at any time will ever make any sense to you or anyone else that could possibly look at them with a shred of rational thought.  Just try to get used to that concept, it will repeat itself over and over.

@Michael - I believe she is giving her absolute cell count, not a percentage.

So she went from 200 (presumably out of 200, i.e. 100%) to 2 out of 200 or 1%

rct,

You cracked me up with that one! So, true!
