TKI Resistance-Why?

Any studies and/or data regarding TKI resistance? I developed a resistance to Gleevec after 18 mos. losing my CCyr and also was resistant to Sprycel after 6 mos. So, now I am awaiting results of a mutation test.What are reasons for being resistant to TKI`S? Is it drinking too much alcohol? Is it homeopathic approaches such as taking flaxseed, turmeric, etc. or is it one`s diet? I`m in good health running 3-4 times a week and my Wbc`s are at 7;Hgb at 14 and Platelets are about 100!Any definitive research on WHY some of of us are resistant?I am soooo frustrated, but I am going to a CML specialist soon and hope I finally meet a TKI that works.Any help would be greatly appreciated, but PLEASE don`t cut my thread-OK?

Oh, I also wanted to add:Do the metabolic rates differ insofar as absorbing any and all TKI`S? would a blood test show the concentration ? Wouldn`t that be clear evidence as to the efficacy of the drug if little is being assimilated in the first place?/

Jlegakis I just wanted to let you know that I so feel your pain. I've had a similar issue - responded well to Gleevec for 9 Mo then PCR shot up, same with sprycel. I'm 2.5 years post diagnosis and just achieved my lowest PCR 4.0% IS. It's so frustrating when you're doing everything you can - taking meds religiously - and still aren't meeting milestones. No significant mutations, no apparent reason, just bad luck?
I just wanted to let you know that you're not alone and wish you the best of luck. Hopefully one of the other TKIs will work well for you.

In the world, there are new mutation tests. For example:

If no mutation is found, maybe a switch to Bosulif and if that doesn't work, try to get into the ABL001 trial.

PCR 53%, 41%, 1.69%, 5.63% (Mutation test negative) September 2013
Sprycel 100 mg
PCR 1.1%, 0.2%, 0.2%, 0.6%, .09%, .06%, PCRU June 2015, PCRU Sept 2015, PCRU Dec 2015, 0.042% Mar 2016, 0.122% April 2016, 0.19% June 2016, 0.176% July 2016, .052% Sept 2016, .031% Nov 2016
Diagnosed Basel Skin Cancer December 2012, October 2014 (All cancer removed)
Diagnosed Melanoma April 2015 (All cancer removed)
