I was diganosed in May 2013 at age 60 and immediately started on Tasigna 600mg/day. I reached MMR at 22 months but due to the continuing fatigue and new onset of metabolic changes, my Hematologist(s) suggest I change to a new TKI. One suggests Gleevec sighting 13 years of safety, while the other supports Bosulif due to the fatigue factor associated with Gleevec. Has anyone had any experiences with this change of TKI's? Again, fatigue is my main complaint. I am currently off meds for a break while checking BCR-ABL monthly.

I have used 5 different TKIs over the last 6 years. I have been taking Bosulif since February and have had the least side effects.
I had not reached MMR on any of the other 4.

One note of caution. I started Bosulif on a full dose of 500mg and after 2 weeks I had a bad reaction that put me in the hospital for 5 days.

I took a 6-week break and resumed at 300 (200 morning / 100 evening) and reached MMR after 3 months and have had minimal side effects over 7 months.

I've also been on several TKIs and am currently on Bosulif. It has proven to have the least side effects for me too. As far as fatigue though, it's still there. Whether it's the CML or the Bosulif, I don't know. Could be both. I do know that when I've had to take drug breaks, the fatigue was less. I should mention that I also am cytopenic, so that's another layer of fatigue.

You say you're changing your TKI because of fatigue and hopefully you've had test to determined if there are any other contributing factors. I hope you're able to find relief with whatever new TKI you try.
