Explanation of PCRU??

Ok - This sounds like a dumb question, but I'm going to ask anyway.  How does PCRU show up on your test results?  Will it actually say 0% or undetectable, or is it a certain log reduction like 4 or 5?  All this stuff is so confusing even after 2 years.  Here are my results if that helps at all (I don't have the results from diagnosis):

     8/5/11 - diagnosed

     12/27/11 - .37% - BCR-ABL Copies 63

     4/9/12 - .035% - BCR-ABL Copies 42 - Achieved MMR

     7/2/12 - .028%- BCR-ABL Copies 18

     9/28/12 - .015% - BCR-ABL Copies 2

     1/15/13 - .0043% - BCR-ABL Copies 3

     4/15/13 - .0099% - BCR-ABL Copies 11

     7/15/13 - .0051% - BCR-ABL Copies 4

Hopefully someone can explain it so my Gleevec brain will understand!!

The "u" stands for undetectable.  So typically the test result will say something like "test results are negative for BCR-ABL".  But it could be expressed a few different ways. 

From an academic standpoint, since the PCR test is not sensitive enough to detect anything below 4.5 log reduction, even if your lab reports below 4.5, most would consider that the same as PCRu aka CMR (Complete Molecular Response)

Hi Cindy:  Like Lucky said the U means undetected.  I am at PCRU, and my results always read 0.000 on the test

results.  I know how confusing it can be when you get down to numbers.  Thats where I always have a problem, so when I go into see my Oncologist I just say Am I  still 0.000 Negative ,  If you do not understand something, just ask all the questions you want when you go to your check-ups, or if you get a call to give you results.

Thanks Lucky - I just wasn't sure how sensitive the test was and how low a level it can detect to.  So if I get one or two more zeros in there, I can say I'm CMR instead of MMR.  Thank you!!

Thanks Susan - I'll just keep waiting for my "0.00".  I was hoping to have it by now, but I know MMR is the most a lot of people ever get so I'm satisfied with that for now.  Just have to wait and see!  Thanks for your response!

I am curious what your PCR report says in the sentence where it tells you the number of "copies".

It's in a table format with the following columns:  Collection Date, %BCR-ABL/ABL (IS), %BCR-ABL/ABL (raw), ABL Copies, BCR-ABL Copies.  Now that I'm looking at it more carefully, it looks like they divide the # of BCR-ABL Copies by the # of ABL copies and that is where they get the %BCR-ABL/ABL (raw) from.  I'm not sure how that translates to the %BCR-ABL/ABL (IS) though.  The numbers from my latest test were IS 0.0051%; Raw 0.0063%; ABL Copies 63795 and BCR-ABL Copies 4.

OK.  BCR-ABL divided by ABL is 4/63795 which is .0063% which is your Raw score.

So on your recent PCR there are 4 BCR-ABL (leukemic gene) for every 63795 copies of ABL (the housekeeping gene). 

I just find the PCR math unusual. 

Anyway, Cindy, you are doing very well.  4+ log reduction by IS.  Congrats.
